Isolation and culture of buffalo embryonic stem cells. A, B In vitro produced hatched blastocysts; inner cell masses mechanically isolated from hatched blastocysts; and seeded on buffalo fetal fibroblast feeder layer (1A, 2A) and putative buffalo embryonic stem cells were formed on 7th day. These putative embryonic stem (ES) cells colonies were passaged 7 times (1D, 4D). The buESCs were maintained in ESC medium which contained BuLIF in the concentration of 40 ng/ml.
